FA proteins (Fanconi Anemia proteins) are a group of proteins crucial for DNA repair and genome stability. Specifically, the FA proteins A, B, C, E, F, G, L, and M form the FA core complex, which functions as a nuclear E3 ubiquitin ligase. This complex plays a vital role in the Fanconi Anemia (FA) pathway by mediating the monoubiquitination of two other FA proteins, FANCD2 and FANCI, after DNA damage occurs.

Role of FA Proteins in DNA Repair

Here's a breakdown of their function:

  • The FA Core Complex: This complex is composed of multiple FA proteins (A, B, C, E, F, G, L, and M). Its primary function is to act as an E3 ubiquitin ligase. E3 ubiquitin ligases are enzymes that attach ubiquitin, a small protein, to target proteins.
  • Monoubiquitination of FANCD2 and FANCI: Upon DNA damage (particularly interstrand crosslinks), the FA core complex monoubiquitinates FANCD2 and FANCI. Monoubiquitination involves attaching a single ubiquitin molecule to the target protein.
  • Activation of the FA Pathway: This monoubiquitination event is critical for activating the entire Fanconi Anemia pathway. The monoubiquitinated FANCD2/FANCI complex localizes to DNA damage sites and recruits other proteins involved in DNA repair, specifically homologous recombination.

Consequences of FA Protein Deficiency

Mutations or loss of any component within the FA core complex disrupts its function and prevents the monoubiquitination of FANCD2 and FANCI. This, in turn, leads to a defective FA pathway and an inability to properly repair DNA damage. Individuals with Fanconi Anemia have mutations in one or more of these FA genes, leading to:

  • Bone Marrow Failure: Due to impaired DNA repair in blood stem cells.
  • Increased Cancer Risk: A higher susceptibility to developing certain cancers, such as leukemia and squamous cell carcinomas.
  • Developmental Abnormalities: Physical abnormalities can occur due to the role of the FA pathway during development.

In essence, FA proteins are critical components of a vital DNA repair pathway, and their dysfunction leads to severe health consequences.
