The rabbit eye test, also known as the Draize test, is an acute toxicity test used to assess the potential of a substance to cause eye irritation or damage.

Draize Test Explained

The Draize test involves applying a test substance to one eye of an albino rabbit, while the other eye serves as a control. Researchers then observe and record any adverse effects, such as redness, swelling, discharge, or clouding of the cornea, over a period of time (typically up to 14 days). The severity of the effects is scored using a standardized scoring system.

Purpose of the Draize Test

Originally developed to evaluate the safety of cosmetics, the Draize test has been used to assess a wide range of substances, including:

  • Cosmetics: Determining if ingredients in makeup, skincare, and hair products cause eye irritation.
  • Pharmaceuticals: Evaluating the potential eye irritancy of drugs and medications.

Criticisms and Alternatives

The Draize test has faced significant criticism due to ethical concerns regarding the use of animals, the subjective nature of the scoring, and the variability of results. Consequently, there is growing interest in and development of alternative testing methods, including:

  • In vitro tests: Using cell cultures or tissue models to assess eye irritation potential.
  • Computational models: Predicting eye irritation based on the chemical structure and properties of a substance.
  • Human volunteer studies: Under carefully controlled conditions, testing substances on human volunteers to assess eye irritation.

While the Draize test is still used in some situations, particularly when required by regulatory agencies, the trend is toward replacing it with more humane and scientifically advanced alternatives.
